        The Human Interleukin 1 receptor antagonist (IL1RA/IL1RN) ELISA kit is designed to specifically detect the IL1RN levels in human serum, plasma, cell culture supernates, or tissue homogenates. This assay employs the sandwich ELISA technique in which IL1RN in the sample is sandwiched between the pre-coated anti-human IL1RN antibody and Biotin-labeled IL1RN antibody and the Ag-Ab-Ag complex is labeled by the HRP-avidin and then develops color reaction after the addition of the TMB substrate solution. The color intensity can be measured by a microplate reader at 450 nm. The kit has been validated with a broad detection range (78 pg/mL-5000 pg/mL), high sensitivity, excellent specificity, precision low than 10%, high recovery, and consistency between batches.

        IL1RN is an agent that competitively inhibits IL1 binding to the cell surface IL-1R and its local inflammatory effects. It is generated locally in various tissues in response to infection or inflammation. High levels of IL1RN is found in the circulation secondary to hepatic production as an acute-phase protein. The imbalance between IL-1 and IL1RN may predispose to inflammatory arthritis and arthritis. IL1RN has been demonstrated to involve in the immunosuppressive effects of mesenchymal stem cells (MSCs). MSCs, in IL-1RN-dependent manner, inhibited IL-1α and IL-1β-driven inflammation, elicited anti-inflammatory M2 phenotype, attenuated antigen-presenting properties of dendritic cells (DCs), and increased generation of Tregs enabling generation of the immunosuppressive milieu in inflamed tissue that led to alleviation of Th1/Th17 cell-dependent immune response and promoted enhanced tissues repair and regeneration.
